My default's Opera (mainly because of its unique view-based e-mail client
which I can't manage without), but I also use Firefox and Safari and IE7
and IE6 to check sites.

HTML Validator, of course, with Serial checking.
Firebug does DOM/CSS/JS etc. so I haven't bothered with web developer or
dom inspector. Do they add additional tools?
I've tried ySlow, but I haven't found it much more use than Firebug's
built-in Net view, and I keep asking myself "why slow?" when I use it ;)
Also I sometimes use:
View Source Chart to see the structure of a page, especially when working
with lots of javascript-generated elements (as in Google Maps API).
All-in-One Gestures, 'cause I'm used to gesturing in Opera.
MeasureIt for an in-browser ruler.
